چکیده

The action of enzyme preparations possessing hyaluronidase activity upon a number of substrates other than hyaluronic acid has been studied by Meyer and his colleagues (1940, 1941), by Madinaveitia & Stacey (1944), and by McClean & Hale (1941). The enzymes, for whose purity no claims were made, were prepared from testis, leech, pneumococcus, streptococcus, and Cl. welchii, and the substrates tested included a wide range of natural mucopolysaccharides and glucuronides, and two synthetic ,B-glucosaminides. These studies, which were mainly qualitative, showed that there was a wide variation in the number of substrates which were attacked, depending upon the source of enzyme. Although there is little doubt that in many of these instances enzymes distinct from hyaluronidase were involved, it has been questioned (East, Madinaveitia & Todd, 1941) whether f-glucosaminidase activity is essential to the complete breakdown of hyaluronic acid, and it seemed that this was a point worth resolving.' In the course of repeating, and confirming, some of the experiments of Meyer and his colleagues it was observed that testis enzyme preparations hydrolyzed chondroitin sulphuric acid almost as rapidly as hyaluronic acid, and the kinetics of this action were briefly studied. also in the hope that the enzyme might prove useful in studying the detailed structure of chondroitin sulphuric acid. This particular action of testicular hyaluronidase preparations has been observed independently by Madinaveitia & Stacey (1944), who recorded, however, only the effect on the viscosity of the substrates which they tested.

Hyaluronidase: correlation between biological assay and other methods of assay.

Different workers have at various times sought to correlate the in vivo and in vitro activities of hyaluronidase preparations. Chain & Duthie (1940), drawing attention to the association between diffusion in the tissues and the mucolytic activity of testicular extracts, pointed out that the viscosityreducing and diffusing activities of various enzyme preparations were in good agreement. McClean...

Isolation, Screening and Characterization of Hyaluronidase Producing Bacteria

Hyaluronidase has a panoramic use in biotechnology processes and therapy due to its therapeutic, pathophysiological, physiological and biological importance. Since much of the preparations of hyaluronidases are from animal source (bovine and ovine testicular sources) with limited sources of microbial origin, that prompted the authors to screen and isolate a new promising bacterial strain ...

Studies on diffusing factors: The hyaluronidase activity of testicular extracts, bacterial culture filtrates and other agents that increase tissue permeability.

AQUEOUS extracts of mammalian testicle contain a factor that dramatically increases the permeability of the tissues to injected fluids [McClean, 1930; 1931; Hoffman & Duran-Reynals, 1931]. This can be demonstrated by the rapid disappearance of the bleb following intracutaneous injection of these extracts and by the rapid spread through a large area of skin of any suitable coloured indicator tha...

Effect of heat and pH on hyaluronidase.

Crude preparations of bovine testicular hyaluronidase (1) depolymerize both hyaluronic acid (HA) and chondroitinsulfuric acid (CSA). Since a series of enzyme preparations with a wide range (loo-fold) of purity shows a constant ratio of activities toward both substrates when examined by a turbidimetric method of assay, the conclusion was drawn that both activities were most likely assignable to ...

The Relationship between Spreading Factor and Hyaluronidase

A comparative study of spreading factor and hyaluronidase in preparations from various sources revealed the following points of similarity and dissimilarity in the two reactions. 1. Similarities: (a) All preparations containing hyaluronidase also produced spreading. (b) Heating at 65 degrees and 100 degrees C. for 30 minutes produced a comparable effect on both reactions. (c) The demonstration ...
